Article: CDH17: The Next Breakthrough in Gastrointestinal Cancer Therapy
Natasha Slepak from KACTUS has recently published an insightful review titled “CDH17: The Next Breakthrough in Gastrointestinal Cancer Therapy”. While identifying specific antigens for solid tumors remains highly challenging, CDH17 targeting is rapidly emerging as a foundational strategy for advanced preclinical research.
What Makes CDH17 Unique in Gastrointestinal Models?
CDH17 (Cadherin 17) is an atypical adhesion molecule that is abnormally highly expressed in gastrointestinal malignancies, such as colorectal, gastric, and pancreatic cancer models. Its unique structure consists of a long extracellular region composed of seven cadherin domains (EC1–EC7), each offering different strategic advantages for antibody design:
-
EC1–EC2: These distal domains are the most exposed regions, making them optimal targets for antibody recognition and maximizing Antibody-Drug Conjugate (ADC) internalization.
-
EC3–EC5: These mid-region domains contribute to structural flexibility and folding, potentially helping researchers differentiate normal conformations from tumor-specific ones.
-
EC6–EC7: These membrane-proximal regions contain an RGD motif that mediates critical integrin recognition and adhesion signaling.
Why is Domain-Specific Design Crucial?
Because CDH17 has a relatively long extracellular region, the specific domain targeted determines the selectivity and functional mechanism of the resulting antibody. By utilizing domain-specific CDH17 targeting, researchers can precisely engineer ADCs, bispecific antibodies, and CAR-T models to block specific cellular interactions while maximizing tumor specificity.
