Exhaustion vs. Dysfunction in CAR-T Cells: Emerging Biomarkers That Are Redefining Their Functional Assessment
Immunostep has recently published an excellent deep-dive into “Exhaustion vs. Dysfunction in CAR-T Cells: Emerging Biomarkers That Are Redefining Their Functional Assessment”
What is the Difference Between Exhaustion and Dysfunction?
While often used interchangeably, these two states represent different phases of a T-cell’s lifecycle in preclinical models:
-
Exhaustion: A progressive loss of effector function due to chronic antigen exposure. This state is often reversible if the immunosuppressive signals are blocked.
-
Dysfunction: A terminal state where the engineered T-cell undergoes irreversible epigenetic changes and can no longer be rescued by standard checkpoint inhibitors.
How Do Biomarkers Redefine Functional Assessment?
To optimize the persistence of engineered cells, researchers must accurately assess their status. Emerging biomarkers are now allowing scientists to pinpoint exactly where a T-cell sits on the exhaustion-to-dysfunction spectrum
