Developing effective vaccines and immunotherapies requires a deep understanding of Germinal Centers (GCs)—the “schools” where B cells learn to produce high-affinity antibodies. Because GC reactions are governed by complex stromal signals and tissue architecture, traditional 2D cultures often fall short. Researchers have now developed modular, PEG-based immune organoids that mimic native lymphoid niches to better predict immune responses across various age states and antigen formats.

Engineering the 3D Immune Microenvironment

By using PEG-4MAL hydrogels incorporated with integrin-binding ligands, this platform provides a tunable 3D matrix that supports primary immune cell survival and differentiation. This system allows scientists to control the “mechanical” and “chemical” cues of the lymph node, revealing how the mode of antigen presentation—whether soluble or membrane-embedded—shapes the magnitude of the immune response.
